羊草种子的休眠生理及提高发芽率的研究 被引量:67

摘要 本研究采用组织培养、高效液相色谱等方法,确定了羊草的休眠类型为抑制物引起的生理性休眠,主要抑制部位是胚乳,关键抑制物之一是脱落酸(ABA);羊草种子的休眠期为4年左右,5至6年达到自然发芽率高峰,此后发芽率开始下降;外源植物激素CAS和6─BA对解除羊草种子的休眠具有促进作用。贮存1年的深度休眠的羊草种子,用低温流水浸泡法处理后能使自然发芽率(土培法,35.0%)提高到88.5%(土培法).不同发芽床上进行的发芽率比较试验表明,土培法和沙培法明显优于纸培法,以土培法的发芽率为最高。 o solve the problem of poor germination in Leymus chinense seeds in cultivation,the re-search work was priorly focused on dormancy.By introducing the methods of tissue cul-ture and high-performance liquid chromatography,etc,it was confirmed that the dor-mant state was imposed by inhibitors.The inhibitory site was endosperm,and ABAwas the main inhibitor.It was also determined that the duration of dormancy for L。chinense seeds was about 4 years. The peak of natural germination appeared in the 5thor 6th year after harvest,and then the persentage of germination tended to decrease.The exogenous hormones(GA_3,6─BA),showed positive effects tor breaking dor-mancy. The percentage of germinaton in L.chinense seeds treated by cold flowing wa-ter changed markedly.Germination percentage of deep dormant seeds preserved for lyear increased from 35.0%(control)to 88.5%in soil culture. The relationship be-tween the seed preservation period,the duration of cold flowing water treatment andthe percentage of germination in seeds,and the possible mechanism of cold flowing wa-ter treatment for improving germination rate were discussed.The comparison of naturalgermination rates from 3 different germination beds showed that the percentage of ger-mination was the highest in soil culture and higher in sand culture than in paper culture.
