摘要 目的:探讨黄芪-壳聚糖/聚乳酸(AP-C/PLA)、壳聚糖/聚乳酸(C/PLA)多孔支架材料的细胞相容性,为牙周骨组织工程生物材料的选择提供依据。方法:将诱导的犬骨髓基质细胞(BMSCs)分别与AP-C/PLA、C/PLA支架联合体外培养。5d取材,进行形态学观察,测定两种材料的吸附率,放免法检测骨钙素的分泌情况。结果:BMSCs在两组材料上能贴附、生长, AP-C/PLA上贴附的细胞数量、细胞吸附率、骨钙素高于C/PLA。结论:AP-C/PLA、C/PLA具有良好的细胞相容性,AP-C/PLA较好,且可能成为前景良好的牙周骨组织工程生物材料。 Objective: To investigate cellular compatibility of the astragalus polysaccharides- chitosan/ polylactic acid(AP-C/PLA) and chitosan/ polylactic acid(C/PLA) with bone marrow stromal cells(BMSC s) in vitro. Methods: Dog BMSC s (induced by 50 mg/L vitamine C, 10 -8 mol/L dexamethasone, 10 mmol/L β-sodium glycerylphosphate)were cultured on AP-C/PLA, C/PLA respectively.5 days after set up, BMSC s attachment and morphology were observed with light microscope and combining rates was detected. The concentration of osteocalin were detected with radio-immunological method. Results: On day 5, BMSC s on AP-C/PLA was significantly more than that on C/PLA. The cell-combining rates and the concentration of osteocalin of the cells on AP-C/PLA were significantly higher than those on C/PLA. Conclusion: Both AP-C/PLA, and C/PLA have good cellular compatibility without cytotoxic. AP-C/PLA is superior than C/PLA, which may be used as a scaffold material for periodontal tissue engineering.
