摘要
目的:探讨细粒棘球绦虫Eg95重组蛋白诱导小鼠的免疫应答效果。方法:设立Eg95重组蛋白免疫组(rEg95组)和对照组(NS组)小鼠分别注射Eg95重组蛋白、福氏佐剂和生理盐水,收集各组血清用酶联免疫吸附(ELISA法)检测IgG水平;采集脾细胞用四甲基偶氮唑盐试验(MTT法)检测免疫小鼠的淋巴细胞增殖反应。结果:Eg95重组蛋白免疫小鼠在第2次免疫后开始检测到抗Eg95抗原的IgG,并随着免疫次数的增多,血清抗体效价升高,在第1次免疫后第10周时,免疫抗体滴度可达到1∶25600。在第4次免疫后,进行淋巴细胞转化实验,MTT法检测证实Eg95重组蛋白免疫的小鼠,其脾细胞可在体外被特异性刺激增生。结论:细粒棘球绦虫Eg95重组蛋白可诱发小鼠产生特异的体液免疫和细胞免疫应答。
Objective: To observe the immunological effect of the recombinant Eg95 proteins in mice. Methods: Anti-Eg95 IgG in murine serum were determined by ELISA. The proliferation activity of the spleen lymphocytes was tested using MTT assay. Results: The specific IgG was induced during the 2^(nd) week and continued to increase until week 10. The specific antibody titers were 1∶25 600 in murine serum immunized by the recombinant Eg95 proteins. There was a significant difference of IgG level between immunized group by the recombinant Eg95 proteins and FCA control In the proliferation response of the spleen lymphocytes, the stimulation index(SI) in the recombinant Eg95 proteins group was higher than that of FCA and NS groups (P<0.01). Conclusion: Eg95 DNA vaccine can induce significant cellular and humoral immune response in mice.
