摘要
针对核桃试管芽苗继代培养玻璃化现象严重、试管芽苗易黄化或老化、继代增殖率低等问题, 本试验在固体培养条件下, 研究了不同培养基、pH值、培养温度及继代间隔时间等因子对核桃试管芽苗继代培养的影响。结果表明: ①新改良DKW培养基上的核桃试管芽苗较改良DKW培养基生长健壮, 增殖率高; ②核桃试管芽苗继代增殖的最适pH值为6 .2; ③变温培养(光照期25℃±3℃, 黑暗期18℃±2℃) 较恒温培养(25℃±3℃) 适于核桃试管芽苗的继代增殖; ④继代培养间隔20d, 试管芽苗生长健壮, 萌发新梢较多; ⑤不同激素组合中, 以BA05 +KT1. 0 +IBA0 001组合效果最好。核桃试管芽苗继代培养的最佳培养条件为: 芽苗在新改良DKW+BA05 +KT1. 0 +IBA0 001 +琼脂7g/L+蔗糖30g/L培养基上变温培养20d。
The effects of medium ,value of pH, culture temperature and subculture days on growth and proliferation of walnut shoots in vitro were studied. The results showed that:①the shoots in vitro cultured on newly modified DKW growed vigorously and proliferated quickly;②the optimized value of pH for subculture proliferation of shoots in vitro shoots was 6.2;③the subculture proliferation of the shoots in vitro cultured in variable temperature(light period 25℃±3℃,darkness period 18℃±2℃) was better than that in invariable temperature(25℃±3℃);④the shoots in vitro subcultured for 20 days were vigorous;⑤the combination of BA 05+KT 1.0+IBA 0.00 produced the best result among different hormone combinations. Therefore, in subculture of the shoots in vitro, the optimal culture medium was newly modified DKW+ BA 05+KT 1.0+IBA 0.001+ Agar 7g/L+Sugar 30 g/L and cultured for 20 days in variable temperature.
