摘要
[目的]观察葛根总黄酮对神经肽Y(NPY)诱导血管平滑肌细胞(VSMC)增殖的影响。[方法]建立体外培养VSMC 体系,实验分为4组,分别为正常对照组(加无血清的DMEM培养液)、模型组(加NPY条件培养液)、正常+葛根总黄酮组(即正常+中药组,加葛根总黄酮条件培养液)和模型+葛根总黄酮组(即模型+中药组,加NPY条件培养液和葛根总黄酮条件培养液),采用荧光免疫组化定量技术检测各组VSMC中的细胞核增殖抗原(PCNA)、血小板衍生生长因子(PDGF)和原癌基因C-myc表达平均荧光强度。[结果]模型组PCNA、PDGF和C-myc表达明显高于正常对照组(P<0.01); 正常+中药组3项指标均降低,与正常对照组比较具有显著性差异(P<0.01);模型+中药组3项指标比模型组降低(P <0.01)。[结论]葛根总黄酮对正常VSMC增殖有一定抑制作用,对NPY诱导VSMC增殖具有拮抗作用,其治疗心血管疾病的作用可能与后者有关。
[Objective] To observe the effect of Pueraria Lobata isoflavones (PLI) on neuropeptide-Y-induced (NPY) vascular smooth muscle cell (VSMC) proliferation. [Methods] Experiment system for VSMC was set up as 4 groups: control group (with DME culture fluid, free of blood serum; group A), model group (with NPY culture fluid; group B), control + PLI group (with PLI culture fluid; group C) and model + PLI group (with NPY and PLI culture fluid; group E). Quantitative fluoroimmunohistochemistry techniques were applied to examine the mean fluorescent values of proliferating cell nuclear antigen (PCNA), platelet derived growth factor (PDGF) and C-myc expression. [Results] The levels of PCNA, PDGF and C-myc expression in model group were higher than control group (P <0.01); those in control + PLI group were decreased, having a significant difference compared with control group ( P < 0.01) ; those in model + PLI group were decreased (P < 0.01 compared with model group) . [Conclusion] PLI may inhibit the VSMC proliferation and counteract the NPY-induced VSMC proliferation. The effect of PLI in the treatment of cardiovascular diseases may be related to its effect on NPY-induced VSMC proliferation.
