颗粒细胞制备单层细胞时其浓度对牛早期胚胎体外发育的影响 被引量:12

摘要 应用颗粒细胞制备单层细胞时,将其浓度分为8组:64×106、32×106、16×106、8×106、4×106、2×106、0.5×106、0个/mL,观察了不同浓度颗粒细胞对体外受精早期胚胎体外发育的影响。试验结果表明,制备单层细胞的适宜颗粒细胞浓度为(0.5~32)×106个/mL,其中16×106个/mL浓度组的囊胚率为42.3±8.0%,明显高于对照组(22.7±8.3%)及64×106个/mL浓度组(25.2±12.O%)(P<0.01)。就发育速度和胚胎质量而言,该组胚胎的发育较快且质量较好,而且标准差反映批与批之间结果较为稳定,是理想的做单层浓度。 The objective of this experiment was to find out the optimum concentra-tion of granulosa cell in preparing granulosa cell monolayers(GCM ) for the culture of early bovine embrvos derived from in vitro maturation (IVM)and in vitro fertilization (IVF). Eight treatments were arranged according to the concentrations of granulosa cell in preparing GCM:64、32、16、8、4、2、0.5、0×106cell/mL, The result demonstrated that:(1) GCM was necessary in IVC, the control had the lowest blastocysts(BL)(22.7± 8.3%)and hatched-BL(28. 4 ±20.5%)rates which were significantly different from the other treatments(P<0. 01);(2)The optimum GCM concentration was 0.5~32 × 106 cell/mL, there were no significant differences among these concentrations in BL and hatched-BL rates (P<0. 05);(3)16 × 106 cell/mL was the best GCM concentration for obtaining more BL, especially at day 7. Embryos in this group developed fast and in good shape ,and results from batch to batch were rather stable。
