黄蜀葵花总黄酮药理性预适应对家兔心肌缺血再灌注损伤的保护作用 被引量:19

Protective effect of pharmacological preconditioning of total flavone of Abelmoschl manihot on ischemical reper-fusion injury myocardium in rabbits

摘要 目的采用心肌缺血预适应实验模型,观察黄蜀葵花总黄酮(total flavone of Abelmoschl manihot,TFA)对家免心肌缺血再灌注损伤是否具有预适应样保护作用。方法采用持续静脉推注TFA模拟预适应的实验方法,观察家免冠状动脉左旋支经结扎、再灌后,其心律失常发生率、心肌组织中LD含量和LDH活力的变化,以及心肌组织中P53,bcl-2表达的情况。结果TFA (16,8,4mg·kg-1)可不同程度降低心律失常的发生率、抑制心肌组织中LD的生成和LDH的漏出、上调bcl-2 mRNA的表达强度,同时下调P53 mRNA的表达水平。结论TFA药理性预适应对家免心肌缺血再灌注损伤具有保护作用。此作用可能与改善心肌能量代谢、抑制心肌细胞凋亡密切相关。 OBJECTIVE: To observe the protective effect of total flavone of Abelmoschl manihot (TFA) on myocardial ischemical reperfusion injury (MIRI) in rabbits. METHODS: Before ischemia reperfusion, the different doses of TFA were perfused intravenously for 5 minutes, followed by a 10 minutes break. The incidence rate of cardiac arrhythmia, the content of lactic acid (LD) and the activity of lactate dehydrogenase (LDH) in myocardial homogenate, and also the apoptosis following ischemia and reperfusion were observed. RESULTS: TFA(16,8,4 mg&middotkg-1)decreased the incidence rate of cardiac arrhythmia, significantly reduced the content of LD in myocardial homogenate, decreased the transudation of LDH from myocardial cells. TFA at the same doses down-regulated the expression of P53, up-regulated the expression of bcl-2. CONCLUSION: Total flavone of Abelmoschl manihot-PP played a protective action on myocardial ischemic reperfusion injury in rabbits by ameliorating the myocardium metabolism and inhibiting apoptosis.
