脱细胞异体神经材料复合雪旺细胞的体外三维构建的活性研究

摘要 目的探讨雪旺细胞在AECM支架材料上体外三维培养的生长活性。方法将培养的雪旺细胞悬液接种在支架材料上,用MTT法、倒置相差显微镜、流式细胞仪、扫描电镜、RT-PCR、测定雪旺细胞的活性。结果AECM上的雪旺细胞各项指标与正常培养的雪旺细胞活性比较,差异无显著意义(P>0.05),实验组雪旺细胞DNA无损伤,且能合成分泌GDNF。结论AECM适于构建具有雪旺细胞活性的组织工程化人工神经。 Objective To detect the activity of SC cultured three-dimensionally on AECM in vitro. Methods Primary cultured SC were used to seed AECM. Then,the activity of SC was examined by methods of MTT、FLC、SEM、STM、RT-PCR analysis. Results There was no difference between the activity of SC cultured on AECM and those of control cells(P>0.05), and the DNA of the former was not damaged. Conclusion AECM seeds with SC was suitable to construct tissue-engineered bioartificial nerve.
