摘要
目的探讨创建大月龄胎儿同种带瓣血管组织库的可行性。方法取脑死亡胎儿15例,胎龄28~40周,体重1500~2600g,热缺血时间<150分钟。其中带瓣主动脉15个,带瓣肺动脉8个。严格无菌操作,-196℃液氮长期保存,组织学观察冷冻前后血管大体形态、内皮细胞及超微结构的变化。结果冷冻前后血管瓣膜无细菌污染,动脉壁支架结构完整,血管瓣膜内皮细胞部分脱落。结论深低温冷冻保存的胎儿同种血管瓣膜结构完整,创立大月龄胎儿带瓣血管组织库切实可行。
Objective To investigate the feasibility of establishing homograft valved conduits bank from late-pregnancy fetus.Method The donor pool were obtained from 15 healthy brain-dead fetus whose gestational ages ranged from 28 to 40 weeks.The warm ischemia times (WITs) were limited within 150 minitues.These homografts were obtained and clipped under stern sterility including 15 valved aortas and 8 valved pulmonary arteries.The homografts were preserved in -196℃ liquid nitrogen for a long time.The morphology of arteries were observed by optic microscope and electron microscope.Result All the results of homograft bacterial cultivation were negative.The optic microscopical pictures of arteries showed elementary structure integrity after thawing.The electron microscopical pictures showed the losing of partial vascular endothelium.Conclusion The structures of fetal homografts valved conduits after cryopreservation remain integrity.It is really possible to establishing homograft valved conduits bank from late-pregnancy fetus.
