摘要
目的探讨喉癌组织中3种膜型基质金属蛋白酶(MT-MMPs)的表达与喉癌浸润转移的关系。方法采用逆转录-聚合酶链反应(RT-PCR)的方法检测24例喉癌组织和周围正常粘膜中3种膜型基质金属蛋白酶(MT1,MT2和MT3-MMP)的mRNA的表达情况,并将其结果与临床及病理学资料进行统计学分析。结果喉癌中MT1,MT2和MT3,-MMP的表达显著高于癌旁组织(P<0.01),并与喉癌的侵袭深度密切相关(P<0.05),而与原发部位和病理分级无明显相关(P>0.05)。喉癌伴淋巴结转移者MT1-MMP的表达明显高于无淋巴结转移者(P<0.05)。结论膜型基质金属蛋白酶在喉癌的发展过程起重要作用,MT1-MMP可作为判断喉癌浸润和转移潜能的可靠指标。早期阻断MT1,MT2和MT3-MMP的表达,将可能对预防喉癌浸润转移有重要作用。
Objective To investigate the correlation of expressions of MT1 , MT2 and MT3-MMP (matrix metalloproteinase-1 ,-2, and-3) with the invasion and metastases in laryngeal cancer. Methods Reverse transcription-polymerase chain reaction (RT-PCR) was used to examine the mRNA levels of MT, , MT2 and MT3-MMP in 24 cases of laryngeal cancer. Results The expressions of MT1 , MT2 and MT3-MMP were significantly higher in laryngeal cancer tissues than those in para-tumorous tissues and had a close relationship with invasive depth. But no significantly different expressions of three MT-MMPs were found in different primary location and different histological grades of laryngeal cancer. The expression of MT1-MMP was obviously higher in patients with nodal metastases than that in patients without nodal metastases. Conclusion MT1 , MT2 and MT3-MMP play an important role in the progression of laryngeal cancer, and MT1-MMP may serve as a reliable maker in estimating invasive and metastatic potency of laryngeal cancer. Suppressing expressions of MT1 , MT2 and MT3-MMP early may inhibit the invasion and metastases of laryngeal cancer.
