摘要
以冬枣和酸枣组培苗丛芽和茎段为试材,以秋水仙素为诱变剂,研究了预培养、暗培养、二甲基亚砜和活性炭对秋水仙素诱导多倍体效果的影响。结果表明:将诱变处理的组培苗丛芽和茎段进行4d的预培养、15d的暗培养,以及在培养基中添加1 5g/L的活性炭或1 0%二甲基亚砜均可有效提高诱变频率,茎段诱变率最高达26 67%,丛芽诱变率最高达36 67%。
The shoot segments and cluster buds of 'Dongzao'(Ziziphus jujuba Mill.) plantlets and sour jujube (Z.acidojujuba C.Y. Cheng et. M.J.Liu) plantlets were used as materials, and colchicines was used as inductor. The effects of preculture, dark culture, active carbon and dimethy1 sulphoxide on in vitro polyploid inducing with colchicine were studied. The results indicated that induction efficiency can be improved significantly by treating the shoot segments and cluster buds with 4 d preculture, 15 d dark-culture, 1.5 g/L active carbon, and 1.0% dimethy1 sulphoxide respectively. The highest induction rate of shoot segments was 26.67% and the highest induction rate of cluster buds was 36.67%.
