灰树花多糖PGF-2的理化性质及化学结构的初步表征 被引量:8

摘要 本文利用仪器分析技术对灰树花多糖级分PGF-2的理化性质和化学结构进行了研究。灰树花粗多糖PGF经DEAE—Sephadex A-25柱层析分离得到一种多糖级分为糖蛋白质缀合物PGF-2,其多糖含量95.4%,纸层析及Sephadex G-200凝胶柱层析证实PGF-2为均一多糖;凝胶渗透色谱测定其数均分子量Mn为118 803 Dal,重均分子量Mw为119 612 Dal;经气相色谱分析PGF-2的糖基由葡萄糖、甘露糖和半乳糖组成,摩尔比为1:2.35:1.22;氨基酸分析结果表明PGF-2的蛋白质由16种氨基酸组成。红外光谱与核磁共振谱证实PGF-2主要存在α型糖苷键。由β-消除反应推断PGF-2中多糖与氨基酸的连接方式以-O-Ser连接。 The physical and chemical properties and chemical structure of a polysaccharide fraction, PGF-2, from Grifola frondosa were studied mainly by instrumental analysis . The polysaccharide fraction (PGF-2) was prepared from crude polysaccharide (PGF), using DEAE-Sephadex A-25 chrorriatography. PGF-2 was a glycoprotein. The polysaccharide content was 95.4 % and the protein content was 2.25 %. The sugar part of PGF-2 was non-starch neutral sugar. PGF-2 showed to be homogenous by paper chromatography and Sephadex G-200 chromatography. Its numeral average molecular weight was 118 803 Dal and weight average molecular weight was 119 612 Dal by gel permeation chromatography. PGF-2 was composed of glucose, mannose and galactose with the molar ratio of 1:2.35:1.22 and 16 kinds of amino acids by GC and HPLC analysis. IR and NMR illustrated that PGF-2 mainly contained a -glucosidic bonds.β -elimination reaction showed that the linkage between sugars and amino acid was the form of -O-Ser.
