川芎嗪注射剂对N-甲基-N-亚硝脲引起大鼠视网膜损伤的拯救效应

摘要 目的:观察川芎嗪注射剂对N-甲基-N-亚硝脲诱导SD大鼠光感受器细胞凋亡的保护作用。方法:2003-03/09在中山大学中山医学院实验动物中心,于雌性SD大鼠生后47d,分别经腹腔注射小、中及大剂量的川芎嗪注射剂,1次/d,并于生后50d腹腔注射N-甲基-N-亚硝脲60mg/kg。在N-甲基-N-亚硝脲处理不同时间后处死动物,取眼球。通过dUTP缺口末端标记试剂盒检测光感受器细胞凋亡;视网膜形态学分析测量周边视网膜的总厚度;反转录聚合酶链反应法检测基因c-fos的表达。结果:N-甲基-N-亚硝脲作用24h后,模型组周边视网膜的光感受器细胞凋亡指数为38.0%;川芎嗪注射剂组分别为30.0%,15.6%和8.4%。作用7d后,模型组周边视网膜的总厚度为37μm;川芎嗪注射剂组分别为41,66和85μm。川芎嗪注射剂可抑制N-甲基-N-亚硝脲诱导的即早基因c-fos的表达。结论:川芎嗪注射剂通过下调基因c-fos的表达,抑制光感受器细胞凋亡,从而部分地保护N-甲基-N-亚硝脲引起的视网膜损伤。 AIM:To observe the protective effect of liqustrazine injection on N-methyl-N-nitrosourea(MNU)-induced apoptosis of photoreceptor cells in rats. METHODS:Form March to September in 2003,low,middle and high doses of liqustrazine injections were injected intraperitoneally into 47-day female SD rats once a day and a single intraperitoneal injection of 60 mg/kg MNU was given to rats after 3 days in animal center of Sun Yat-sen Medical College of Sun Yat-sen University.At different intervals after MNU treatment,the animals were sacrificed to get their eyeballs.The apoptotic index of photoreceptor cells was calculated by TUNEL labeling,peripheral retinal thickness was evaluated with retinal morphology,and the expression of c-fos gene was detected by reverse transcription-polymerse chain reaction technique. RESULTS:In the peripheral retina of MNU-treated rats,the apoptotic index at 24 hours after MNU treatment was 38.0 %,and it was 30.0 %,15.6 %and 8.4 %in liqustrazine injection treated rats respectively.Total peripheral retinal thickness at 7 days after MNU treatment was 37 μm in model group,and 41,66 and 85 μm in liqustrazine injection groups respectively.Compared with MNU-treated rats,the gene expression of c-fos was time-dependently down-regulated in liqustrazine-treated group. CONCLUSION:Liqustrazine injection partially protects against MNU-induced retinal damage by down-modulating the expression of c-fos gene to inhibit apoptosis of photorecptor cells.
