斑叶竹节秋海棠微繁殖的研究 被引量:3

摘要 以叶片为外植体,在附加0.4mg/l 6—BA和0.1mg/l NAA的MS培养基上诱导长芽。在1/2MS或附加0.1mg/l IAA的1/2 B_5培养基上诱导出根。对提高繁殖率、降低成本和试管苗移栽成活等做了系统的研究。结果表明。在芽的诱导中,6—BA和NAA起关键的作用,碳源以化学纯蔗糖较好,水质以蒸馏水最好,去离子水和自来水效果很差。但在诱导生根中,市售白糖可代替化学纯蔗糖,自来水可代替蒸馏水。培养物分割转移次数对苗的增殖有明显的影响,次数过少,增殖率明显下降。生根培养基两次利用和在室内自然光下生根对批量生产都有重要的实用意义。现已小批量生产。 The present paper deals with study on the micropropagation of Begonia macuata Radd. The buds were produced when young leaf segments as explants were cultured on MS agar medium, supplemented with 0.4 rag/1 6-BA and 0.1 mg/1 NAA, which then grow into shoots, when the Shoots which reached 3-4cm in height were cut off and cultured in 1/2MS agar medium or 1/2B_5 agar medium supplemented with 0.1mg/l IAA, roots were produced. During the inducing buds, 6-BA and NAA in the medium play a determinative role; chemical pure sucrose is better than commercial white sugar as a carbon source; as far as the quality of water is concerned, distilled water is the best, while tap water and deionized water are bad for the culture effect. But during the inducing rooting, chemical pure Sucrose may be replaced by commercial white Sugar, and distilled water by tap water. Effect of frequency of cutting apart or transfer of the cultures on multilplication of shoot is obvious; if the frequency is too few, the multilplication rate markedly drops. Both twice times use of the rooting medium and the rooting under sunlight within room may be applied to the mass production of B. macuiata Radd., because they are more economical. The sugar content in the rooting medium must be no lessthan 2%, when is 1%, the colour of root changes into black. The rooted plantlets can survive under not subjucted to any hardening before transplantated into pot and the survival rate generally reached above 90%. The transplantated plantlets grow well and bloomed.
