p16 gene expression and clinical significance in human osteosarcoma

摘要 目的:用免疫组织化学LSAB法研究抑癌基因p16在人骨肉瘤中的表达与骨肉瘤预后、分级的关系。方法:30例骨肉瘤新鲜标本,按刘氏分级法分为,,级;三年随访结果死亡13例,存活17例。15例正常对照骨组织。结果:115例骨组织中阳性率73.33%(11/15);30例骨肉瘤组织中阳性率36.67%(11/30),骨肉瘤组较正常骨组p16蛋白染色阳性率和染色强度低(P<0.05);213例死亡组中阳性率6.67%(1/13),17例存活组中阳性率58.82%(10/17)。存活组较死亡组p16蛋白染色阳性率和染色强度高(P<0.01);3p16蛋白的染色阳性率和染色强度与病理分级无关。结论:骨肉瘤的发生与p16基因表达的异常有关;其表达阳性率和强度与病理分级无关,与预后相关,提示p16可作为骨肉瘤的预后判定指标之一。 Objective To study the relationship between p16 gene expression, follow-up and grade of osteosarcoma by LSAB method.Methods Thirty fresh specimens of human osteosarcoma tis- sues were classified into three grades (grade 15 cases, grade 10 cases, grade 5 cases) according to the criteria of Liu. Thirteen cases died and 17 survived in 3 years' follow-up. The control group in- cluded 15 cases of normal osseous tissues.Results1 The positive rate was 73.33% in normal osseous tissues and 36.67% in osteosarcoma tissues. The positive rate and intensity of staining of osteosarco- ma tissues were lower than those in normal osseous tissues (P<0.05).2 One showed weak positive in 13 cases who died and 10 in 17 cases who survived. The positive rate and intensities of staining in survival group were higher than those in death group (P<0.01).3 There was no relation between p16 protein expression (positive rate and intensity of staining) and grade. Conclusion The abnormal expression of p16 gene may play an important role in human osteosarcoma genesis. The expression (positive rate and intensity of staining) of p16 has no relation with histological grade but with relation to follow-up. It is suggested that p16 gene have prognostic value, and the expressed degree of p16 gene can be used to guide clinical therapy.
