摘要
本文报道了杂交水稻12个不同组合幼穗的体细胞胚诱导与同步化的研究结果。结果表明:材料的基因型对其体细胞胚发生有明显的影响,植物激素2,4—D在杂交水稻体细胞胚胎发生与发育中起重要作用,BA则起一定的协调促进作用。通过研究建立了杂交水稻悬浮细胞系与产生大量体细胞胚的培养程序。利用生长素2,4—D结合机械筛选的方法,实现体细胞胚同步率达85%以上。通过组织学观察发现体细胞胚的发生基本来源于胚性愈伤组织表面的细胞,其发育似合子胚,但出现了双子叶形胚和少数畸形胚。
This paper reported the resultes including the somatic embryogenesis and the synchronous control of somatic embryoes from the young ear explant of twelve combination of hybrid rice. The resultes indicated: the genotype of hybrid rice have obvious effect on its somatic embryogenesis , plant hormone 2,4-D have important role in the induction and development of somatic embryoes , BA can control this process in some way. the culture steps to obtain suspension cells and somatic embryoes have been established, the somatic embryo synchronous percentage was above 85 % by using hormone and mechanical sifter. The somatic embryo come mainly from the surface cells of callus, and its development was similar to the zygote embryo,but a few somatic embryoes in culture were abnormal.
