摘要
目的建立对生化汤血清药物化学的初步研究。方法通过建立生化汤及po生化汤后小鼠血清HPLC指纹色谱分析方法,比较生化汤各单味药、给药及未给药大鼠血清的HPLC指纹图谱,同时以保留时间相对偏差△t作为参考来衡量各谱峰的相关性,确定Po生化汤后小鼠血中移行成分及其来源生药。结果生化汤给药后血中出现了9个移行成分,6个为生化汤中所合成分的原型,3个为新产生的代谢产物,其中有一成分相对含量较高。结论保留时间相对偏差△t可在某种程度上为谱峰来源作定性分析。血中移行成分及代谢产物将成为生化汤的体内作用基础,其来源为生化汤中的主要物质,对其进行深入的研究将有助于阐明生化汤的有效部位及作用机制。
OBJECTIVE: To investigate the parameters of serum pharmacochemistry of Shenghua Tang. METHODS: Based on the established HPLC fingerprint analysis, comparison was carried out among the peaks of control serum, serum containing drug, Shenghua Tang and the ingredients of crude drug. Simultaneously, the relative deviation Δt of retention time was utilized as index to evaluate the relation among the fingerprint peaks so as to confirm the original of peaks. RESULTS: Nine compounds were obtained, six of them were the original compounds contained in Shenghua Tang, three of them were metabolites and one of three metablites was found to be high content. CONCLUSION: To some extent, the relative deviation Δt of retention time qualitatively analyze the originals of peaks. The compounds are absorbed into blood and their metabolites are the effective constituents, the original crude drugs are the main ingredient of the prescriptions. Its serum pharmacochemistry should be subject to complete investigation so as to illuminate the pharmacology and active mechanism of Shenghua Tang.
