摘要
目的:为了证明蝎皮对免疫功能的影响并探讨蝎皮中含有的活性成分,通过提取分离蝎皮中的水溶蛋白质和角蛋白质组分,经过体外药理活性筛选,观察其对细胞或体液免疫系统的影响,以期获得具有生物活性的蛋白质组分。方法:采用水提与盐析法获得水溶性总蛋白(ST1),还原法获得总角蛋白(ST2),通过Sephadex G-50凝胶色谱对水溶总蛋白和总角蛋白进行分离,并对获得的各种蛋白组分进行了体外药理筛选。结果:发现蝎皮的蛋白质组分在体外药理实验中显示对T/B淋巴细胞转化具有显著促进作用,但对NK活性无明显作用。结论:蝎皮蛋白提取组分具有一定的免疫调节生物活性,研究结果对蛋白质药物开发提供了基础数据。
Objective: Extracting the total protein from scoipii tegument, investigating its effect on immune system by transformation of T/B cell. Method: Water-soluble protein(STl) was extracted by distilled water method and salting-out method, while keratin(ST2) by deoxi-dization method. Sephadex G-50 was used to isolate the protein. The effects of components isolated by sephadex C-SO on T/B and NK cell were investigated. Result: The protein from scorpii tegument could increase the transformation of T/B cell distinctly in vitro, while no apparent effect on NK cell. Conclusion: Protein from scorpii tegument could modulate the immune system, which may offer a new way for people to find protein drugs.
