摘要
目的研究胃癌(gastriccancer,GC)中环氧合酶-2(cyclooxygenase-2,COX-2)、核因子-κB(nuclearfactorκappaB,NF-κB)的表达规律与病理特征的关系。方法采用免疫组织化学染色法(SABC法)和末端脱氧核苷酸转移酶(TDT)介导的dUTP缺口标记法(TUNEL)检测15例正常胃黏膜(normalgastricmucosa,NGM)和40例GC组织中COX-2、NF-κBp65表达情况。结果GC中COX-2、NF-κBp65的阳性表达分别为67.5%、62.5%,NGM组则为13.3%、6.70%,GC组和NGM组差别有显著性(P<0.01)。COX-2、NF-κBp65在低分化GC中表达显著高于高分化型,且后者还与GC病期有关(P<0.05)。COX-2、NF-κBp65与GC淋巴结转移、血管浸润均密切相关(P<0.05)。结论COX-2、NF-κB的表达水平对评价GC的恶性程度、有无转移和预后有一定参考价值。
[Objective] To study expression of cyclooxygenase-2 (COX-2), nuclear factor-kappa B(NF-κB) in the gastric cancer and their relation with pathological grades and clinical stages. [Methods] Gastric tissue specimens were obtained from 15 cases with normal gastric mucosa (NGM), and 40 with gastric cancer (GC). Expression of COX-2 and NF-κB p65 were detected by immunohistochemistry ( with SABC method ). [Results] Positive rates of COX-2 and NF-κB p65 in GC were 67.5% and 62.5%, 13.3% and 6.7% in NGM respectively. There was a significant difference between GC and NGM(P <0.05). Expression of COX-2 and NF-κB p65 in poorly differentiated cancer was markedly higher than that in well differentiated cancer (P <0.05); furthermore, NF-κB p65 was related to the clinical stages of GC. Expression of COX-2 and NF-κB p65 were all closely correlated with lymphnode,,metastasis and vascular invasion (P <0.05). [Conclusion] Expression levels of COX-2 and NF-κB may be helpful for evaluating malignant degree of GC, lymphnode, metastasis and prognosis.
