核酸检测方法在献血员肝炎筛查中的初步应用 被引量:2

摘要 目的将核酸检测方法应用于献血员肝炎筛查,了解某地区血液安全水平和常规血液筛检中两次酶免检测方式的可靠性。方法44份Murex-abbott抗-HCV阳性标本经科华酶免试剂复检后,使用罗氏COBASAmpliS-creen单独检测HCVRNA;486份来自某地区的合格献血标本采用24份标本混合检测的方法,用罗氏COBASAmpliS-creen检测HBVDNA和HCVRNA。结果44份标本用科华酶免试剂复检,结果只有13份标本呈抗-HCV阳性;3份HCVRNA阳性标本中1份为科华试剂检测抗-HCV阴性。486份合格献血标本进行罗氏COBASAmpliScreen24份混合形式筛检后,发现3例HBVDNA阳性标本,阳性率为0·62%,未发现其他核酸标记物阳性的标本。结论我国血液筛查的HBsAg检测存在漏检的现象,在常规血液筛检中,NAT检测只能作为酶免抗体检测的一种补充手段,如果NAT能取代我国血液二次酶免筛检中的一次,则能在最大程度上保障血液安全。 Objective To dectect hepatitis virus for screening blood donors by using nucleic acid testing(NAT) and survey the security level of blood transfusion in a certain area. Methods 44 anti-HCV positive blood samples and 486 qualified blood samples were collected from a certain area. The HCV-RNA and HBV-DNA of each sample was detected using NAT with COBAS AmpliScreen kits. Results 13 out of 44 anti-HCV positive samples by Murex-abbott kits was negative by Ke-hua kits; 3 blood samples were positive by COBAS AmpliScreen kits.486 qualified blood samples was dectected positive with HBV-DNA by COBAS AmpliScreen kits.with a pasitive rate of 0.62%. Conclusion Some HBV positives may be missed in routine blood screening and NAT is recommended for screening of hepatitis viruses.
