用人胎肝细胞重建SCID鼠人免疫系统 被引量:3

RECONSTITUTION OF HUMAN IMMUNE SYSTEM IN SCID MOUSE WITH INTRAPERITONEAL INJECTION OF FETAL LIVER CELLS

摘要 用人胎肝细胞重建重症联合免疫缺陷鼠(SCID)人免疫系统(SCID-hu)的初步结果。通过免疫组织化学及流式细胞术检测,腹腔内注射胎肝细胞第2周,SCID鼠脾内即可见人淋巴细胞,至第10周仍可测到CD20^+、CD19^+、CD3^+人淋巴细胞;用HBx蛋白重复免疫SCID-hu鼠后第2周,外周血中可测到人源抗HBx抗体(IgG)。提示:人干细胞可在SCID鼠体内存活、分化、成熟,并能对抗原产生特异性的初次及再次免疫反应,腹腔内注射人胎肝细胞是一种较简便、实用的重建SCID-hu的方法,并可用于人源单抗、肿瘤及HIV等病毒感染性疾病的研究;SCID-hu可望解决人源单抗制备中B细胞来源困难的问题。 By intraperitoneal injection of fetal liver cells (FLC,the human immune system (SCID-hu) in severe combined immunodeficiency mouse was reconstituted. Human lymphocytes (CD20,CD19,CD3) could be found in the spleen and peripheral blood from the second to the tenth week after injection. When immunized by HBx protein,human IgG a-gainst HBx protein could be found in peripheral blood of the SCID-hu mouse. These suggest that human lymphoid progenitor and stem cells are able to live,undergo maturation and defferentiation in SCID mouse in the absence of human cytokines. The lymphocytes in the SCID mouse could produce primary and secondary immune response to antigen. SCID-hu could be more easy reconstituted by intraperitoneal injection of FLCs,and might be used in the study of human monoclonal antibodies,tumor and virus infectious diseases.
