摘要
作者合成了一段与人和小鼠干细胞因子(SCF)cDNA能特异性互补的寡核苷酸探针,经放射性同位素标记,与两株人骨髓基质细胞系(HECL、HFCL)和一株小鼠骨髓基质细胞系(TC-1)的RNA进行点杂交。结果表明这三株基质细胞都能表达SCFmRNA。因此,作者利用逆转录-多聚酶链反应技术扩增编码SCF的部分cDNA序列,并将其克隆入pUC18载体。DNA序列分析结果显示作者所克隆的SCFcDNA序列与文献报道的一致。
n oligonucleotide probe matching specificallywith human and mouse stem cell factor (SCF) cDNAwas synthesized, and a dot blot was performed usingthe radiolabelled probe and RNAs extracted from twohuman bone marrow stromal cell lines(HECL, HFCL )and one mouse bone marrow stromal cell line (TC-1).The results showed that these three stromal cell linesexpressed SCF mRNA. By using reverse transcription(RT)-polymerase chain reaction (PCR) technique weamplified a part of the SCF enceding cDNA, andcloned it into pUC18. DNA sequencing results showedthat the SCF cDNA we cloned is identical with that re-ported in literature.
