摘要
应用SPC—A1人肺腺癌细胞系研究了顺铂和卡铂的细胞毒效应及其与人外周血LILAK细胞的联合效应及机制。结果表明:经一定浓度的顺铂或卡铂作用一定时间后,SPC—A1细胞可受到不同程度的杀伤。经1vg/ml的顺铂处理24小时,洗去药物后加入LILK细胞,SPC—A1细胞被明显杀伤,与未经顺铂处理的对照组相比P<0.001;而卡铂在同样条件下却无此效应。由于经顺铂处理的SPC—A1细胞对TNF—2有杀伤敏感性明显增加,推测该因子可能是介导顺铂与LILAK细胞协同效应的机制之一。
The effects of cisplatin(CDDP),carboplatin(CBDCA) , lectin
induced lymphokineactivated killer(LI LAK)cells and their
combination on killing SPC A1, a human lungadenocarcinoma cell line
, Was investigaled. The results showed that after 48h exposure of SPC
Alcells to CDDP or CBDCA , both drugs could kill the target cells in
a dose dependent manner.Pretreatment of the target cells with
CDDP,but not with CBDCA,could markedly increase thesusceptibility of
the tumor cells to lysis by LI LAK cells.Because the susceptibility
of CDDP pretreated target cells to tumor necrosis factor(TNF)was
also enhanced, it was supposed that thisfactor might be one of
important mechanisms in killing CDDp pretreated SPC Al cells bv LI
LAK cells.
