摘要
将马铃薯Y病毒中国分离物(PVY-C)的外壳蛋白(CP)基因,在土壤农杆菌LBA4404的介导下,转化烟草生产品种NC89,获得了6个转基因烟单株系。通过抗性分析发现,6个株系中有一个株系在200μg/mlPVY-C的攻毒下,仍未发病。分子检测发现,转基因植物中抗性产生的程度并不是同PVY-C外壳蛋白的表达水平成正相关。
The coat protein(CP)gene of potato virus Y(Chinese isolate)was transferred into tobaccothrough the mediation of Agrobaacterium tumefaciens LBA4404.The transformed plants wereanalysed by PCR followed by DNA hybridization,Northern and Western blot.The transgenicplants were challenged with PVY-C inoculation.One of the six transgenic lines displayed highresistance at the inoculum dose of up to 200μg/ml of PVY-C. To our knowledge,it is thehighest virus concentration used, compared with other published results.The Western blot analy-sis showed there is no relationship between the amount of the coat protein expressed and theresistance level of the transgenic plants.
