摘要
本研究以革果品种千秋试管苗叶片为试材,接种于MS附加不同浓度BA和NAA激素组合的培养基上,经愈伤组织诱导、不定芽诱导,一步诱导千秋叶片再生不定芽。得到苹果品种千秋叶片-→愈伤组织-→不定芽再生的适宜培养条件为:MS基本培养基附加BA2mg/L,NAA0.2mg/L;黑暗诱导5个星期后在同种培养基中转入光照培养(15001x,每天光照15h),培养室温度为25±2℃,组织学观察表明,不定芽起源为愈伤组织薄壁细胞。本文还研究了激素配比、暗处理时间长短、叶片在植株上着生的节位、叶片在培养基中的接种方式对千秋叶片不定芽再生的影响,及同一培养基中不同苹果品种(千秋、嘎拉、富士)试管苗叶片再生不定芽的能力,并探讨了该技术在苹果育种中的应用前景。
Adventitlous shoots were regenerated from in vitro leaves of apple cultivar Senshu viaorganogenesis.The suitable one-step-way culture included:MS+BA 2mg/L+NAA 0. 2mg/Lmedium culture, 25±2℃ 5 weeks of dark treatment followed by light treatment(1500lx,15h/d)on the same medium for shoot growth.Histological study showed that the adventitiousshoots were from parenchyma cells of leaf callus.Effects of phytohormone concentration,dark treatment,position of leaves on apple plant,placement method of leaves in medium onadventitious shoot regeneration of Senshu were studied. The difference of different applecultivar regenerations(Senshu,Gala,Fuji)in the same medium and the use of this techniquein apple mutation breeding were also discussed.
