摘要
目的观察高+Gx过载作用对猴脾脏组织热休克蛋白-70(HSP-70)表达的影响。方法9只雄性恒河猴,随机分为4组:对照组与实验1、2、3组。经受+Gx作用的强度(或峰值)和时间分别为1Gx、300s;+15Gx、200s;+18Gx、165s;+21Gx、140s。使用专用动物离心机,加载后动物即刻处死、取材、制片、染色。用病理学方法和免疫组化法观察猴脾脏组织学改变和HSP-70表达。结果对照组和实验组脾脏组织大体形态和结构未见明显异常;光镜下见实验各组脾脏有不同程度的充血,尤以高负荷组明显。免疫组化染色显示对照组少量散在的淋巴细胞呈棕色,为HSP-70弱阳性表达;实验组大量广泛的呈深棕色的HSP-70阳性细胞,为强阳性表达。各实验组之间无明显差异。结论高+G过载作用可诱导脾脏淋巴细胞HSP-70表达增强。
Objective To observe the change of expression of heat shock protein 70 (HSP-70) in the spleen of Rhesus macaque under +Gx loads. Methods Nine male Rhesus macaques were randomly divided into control group and 3 experimental groups. Using a special animal centrifuge, control group was exposed to +1 Gx/300 s, and experimental groups were exposed to +15 Gx/200 s, +18 Gx/165 s and +21 Gx/140 s respectively. The animals were terminated immediately after Gx exposure. Spleen tissue was fixed, sectioned and stained for histopathological examination and the expression of HSP-70 in the spleen was detected with immunohistochemical staining. Results As compared with controls, no changes in the gross appearance of the spleen were observed in 3 experimental groups. Microscopically, congestion of the spleen was found in the experimentals, and the intensity of splenic congestion was significantly correlated with the underwent of +Gx loads. Weak expression of HSP-70 was noted in a few scattered lymphocytes of the spleen in control group, while deep brown strong expression of the protein in a great number of cells was found in the experimental groups. No significant difference of the expression of HSP-70 between the experimental groups was found. Conclusions +Gx overload can induce the enhancement of HSP-70 expression in the spleen of macaques.
