摘要
采用连续监测法测定以NADP为辅酶的异柠檬酸脱氢酶(ICD),并对酶促反应条件进行了研究。以Tris-HCl为缓冲体系,酶促反应最适pH为7.5,Mn(2+)为2mmol/L,在最佳条件下测定ICD的线性范围为0~85U/L,批内变异系数2.06%~5.85%,批间变异系数为3.71%~8.72%,100例健康人血清参考范围为2.0~11.2U/L,初步临床应用表明各类肝炎及肝癌均显著升高,ICD/ALT比值对肝癌的鉴别诊断有一定的应用价值。
A kinetic assay of serum isocitrate dehydroenase(ICD) with NADP was reported in this paper. Optimized reagent and react condition were studied.Optimum pH was at 7.5 for Tris-HCl buffer.and Mn(2+) was 2mmol/L.The linear range is from 0 to 85 U/L.Within and between batch CV are 2.06%~5.85% and 3.71%~8.72%. Serum ICD in 100 healthy subjects was 2.0~11.2U/L.The clinical application of the test proved that it is useful to differential diagnosis of hepatic cancer.
