同工酶技术及其在双孢蘑菇选育种中的应用——Ⅰ.双孢蘑菇同工酶标记筛选研究 被引量:16

摘要 采用聚丙烯酰胺凝胶垂直板状电泳和琼脂糖凝胶板状电泳,研究不同生长发育期,不同组织、不同培养基或栽培条件,对双孢蘑菇8种同工酶和可溶性蛋白电泳表型的影响,发现酯酶、多酚氧化酶、细胞色素氧化酶、过氧化物酶、乙醇脱氢酶和可溶性蛋白的主要活性区带存在与否,在营养菌丝生长阶段或子实体发育阶段的相同组织中不受上述各因素的影响,并发现不同类型菌株的电泳表型具有不同的活性区带。把这类活性区带命名为标记区带。把这类活性区带命名为标记区带。以标记区带对同工酶表型进行分类,发现酯酶同工酶是双孢蘑菇生化鉴定或遗传分析的最适标记之一。 The effect of the different growing stages, different tissue and different mediums or cultivated environments on the electrophoretic patterns of eight isozymes and soluble protein from the different strains of Agaricus bisporus were investigated by the polyacrylamide slab gel electrophoresis (PAGE) and agarose gel electrophoresis (AGE). It was found that the appearance of main active bands of Esterase, Polyphenol oxidase, Cytoehrome oxidase, Peroxidase, Alcohol dehydrogenase and soluble protein from the vegetative hyphae or the same tissue of the fruit body were not influenced by the factors mentioned above and found there were different mein active bands in some of the different strains. The main active bands were designed as marker bands. When the isozymic patterns were classed according to the marker bands, it was found that Esterase isozyme is one of the most suitable genetic marker for Agaricus bisporus.
