软骨型蛋白多糖的提纯及生物化学特点的研究

摘要 本文分别用Q-Sepharose柱层析和氯化铯平衡密度梯度离心法从仔猪软骨提纯了软骨型蛋白多糖单体,它在SepharoseCL-2B柱层析上显示了大分子的特点(Kav=0.22).能够与透明质酸结合形成巨大分子的聚合体。提纯的核心蛋白在SDS电泳上显示出单一的银染色区带,分子量为460kDa,与抗人蛋白多糖单克隆抗体(3-B-3)呈强阳性反应.离子交换层析法简便、快速、经济更适合于以蛋白多糖损害为主要特点的软骨疾病的研究。 The large cartilage-specific proteoglycans were purified from pig chondrocyte cultured using method of Q-Sepharose ion exchange chromatography or isopycnic CsCl density gradient centrifugation. The purified proteoglycanm showed the large (90K, Kav=0.22) and small (10%) of hydrodynamic size on Sepharose CL-2B chromatography. Core polypeptide from large proteoglycan (generated by digestion with chondroitinase and keratanase) was purified on Superose 6B chromatography. It showed silverstained single band of 460kDa on SDS polyacylamide gel electrophoresis, and strongly reactivity with an antiserum to human proteoglycan in Western blood. This fast, simple, and low cost method of Q-Sepharose chromatography produces high quality proteoglycans, suitable for studies on proteoglycan in the injured cartilage.
