摘要
联合应用薄层色谱、紫外光谱测定小鼠体内安定。样品制备是在组织匀浆中加入磷酸二氢钠缓冲液(3mol/L),以乙醚振荡提取,离心分离。利用值、紫外吸收峰与谷值、吸收度比等进行安定的特征检识。薄层色谱条件:硅胶G板,分别用苯-丙酮(3:2)及丙酮-苯-氯仿(2.5:4.0:5.0)作为展开剂。定量分析结果表明以A值为定量参数较之以上A值为定量参数有更高的灵敏度;而测定值的偏倚随样品浓度增大而变小;不同介质和不同浓度水平下的回收率不同(60~90%)。小鼠口服安定1h后肝和脑中可明显检出安定,但胃中的安定浓度高得多。
Diazepam in rat tissues was detected by using thin layer chromatography(TLC)and ultraviolet spectrometry(UV).The results of quantitative analysis showed that sensitivity would be higher with A value as the quantitative parameter than with the A value,and the bias would become small as the sample concentration level was high.The recoveries obtained from different sample level were varied in the range of 60%~90%.Diazepam could be detected in rat stomach,liver and brain after being taken for 1h,but the concentrations in stomach were much higher.
