摘要
给大鼠胸膜腔内注射角叉菜胶(10mg·kg-1)复制胸膜炎。粉防己碱(10~80mg·kg-1)于致炎前30min和致炎后4h两次给大鼠ig能明显抑制炎症白细胞磷脂酶A_2(PLA_2)和钙调素活性.并降低胞浆内游离钙浓度:在体外,粉防己碱(20~160μmol·L-1)与白细胞悬液孵育30min亦能明显抑制炎症白细胞PLA_2活性.这种作用可被Ca ̄2+和钙调素提取液逆转。提示粉防己碱降低炎症白细胞PLA_2活性的作用与其拮抗Ca ̄2+、钙调素系统有关。
A pleuritis was induced by an intrapleural injection of carrageenan ( Car, 10 mg· kg-1) in rats. The phospholipase A2 (PLA2) activity, calmodulin activity and cytosolic free calcium concentrationsof neutrophlies from the inflammatory exuate obviously decreased with intragastric administration of tetrandrine( Tet 10~80 mg· kg-1) to rats at 30 min before and 4 h after the injection of Car.Meanwhile.thePLA2 activity of the neutrophiles also decreased when Tet(20~160μmol·L-1) andthe neutrophiles suspension were incubatedtogether for 30 min at 37℃in vitro.The action was reversed in part by calcium and calmodulin extract. These findings suggestthat me inhibition of Tet on me PLA2 activity of neutrophiles may be related to its antagonism on calcium-calmodulin system.
