摘要
目的研究口腔鳞状细胞癌细胞中Toll样受体3(TLR3)的表达及其配体poly(I:C)的生物学作用。方法 RT-PCR和流式细胞术检测口腔鳞状细胞癌细胞系CAL-27和HB中TLR3mRNA和蛋白表达。以100μg/mLpoly(I:C)处理CAL-27,分别于刺激后24、48和72h时点采用MTT比色法测定细胞活力,24h时点采用流式细胞术检测细胞凋亡;并设立空白对照。结果 CAL-27和HB中TLR3mRNA和蛋白均呈高表达。CAL-27经poly(I:C)处理后,各检测时点的细胞活力均显著低于空白对照细胞(P<0.05);细胞凋亡率明显高于空白对照细胞[(16.67±1.202)%vs(7.00±1.155)%](P<0.05)。结论口腔鳞状细胞癌细胞内高表达TLR3,其配体poly(I:C)能诱导口腔鳞状细胞癌细胞凋亡。
