摘要
目的研究小鼠外周血自然杀伤细胞(NK)细胞分化抗原CD69+/NKG2D+检测在保健食品免疫调节作用评价中的应用。方法以左旋咪唑25mg(kg·bw)连续灌胃3d设立左旋咪唑阳性对照;进行小鼠外周血自然杀伤细胞(NK)亚群CD69+/NKG2D+检测和脾NK细胞活性测定试验,观察0.83,1.67g/(kg·bw)剂量组蛋白粉对小鼠NK细胞免疫功能的影响。结果与阴性对照组比较,左旋咪唑对照组和0.83,1.67g/(kg·bw)剂量组蛋白粉剂量组外周血NK细胞CD69-/NKG2D+、CD69+/NKG2D+比值以及脾NK细胞活性均明显升高。结论外周血NK细胞CD69+/NKG2D+检测与传统的NK细胞免疫检测结果有较好一致性,且方法敏感性较好,在保健食品免疫调节功能评价中有较好应用价值。
Objective To apply the peripheral blood NK cell CD69^+/NKG2D^+ in the imunlty function evaluation of health food. Methods To build the positive control through treated with levamisole [ 25 mg/(kg· bw)] by oral feeding for 3d; Peripheral blood NK cell Sub-cluster CD69^+/NKG2D^+ , spleen NK cell activity test were carried out to investigate the effects of protein powder[0.83, 1.67 mg/(kg·bw)] on NK cellular immunity function of mice. Results Compared with the negative control, activity of spleen NK cell and peripheral blood NK cell sub-cluster CD69^-/NKG2D^+ and CD69^+/NKG2D^+ of Levamisole group and [0.83, 1.67 mg/(kg·bw)] group were all significantly increased. Conclusion Levamisole has enhancing effects on the NK cellular immunity function of mice, and can be used as the positive control in the imunity function evaluation of health food;The results of the peripheral blood NK cell CD69^+/NKG2D^+ and the traditional test for activity of NK cell have good coherence, and the former method has good susceptivity and application value in the NK cellular immunity function evaluation of health food.
