摘要
以患丛枝病豫杂一号泡桐叶片、叶柄和茎段为外植体,建立了其体外植株再生系统.结果表明,其叶片、叶柄和幼嫩茎段在MS+NAA 0.3 mg.L-1+6-BA 12 mg.L-1,MS+NAA 0.3 mg.L-1+6-BA 8 mg.L-1和MS+NAA 0.3 mg.L-1+6-BA 12mg.L-1组合培养基上,最高芽诱导率可达47.78%,21.11%和42.22%.此外,不含NAA和IBA的1/2MS培养基可作为患病苗幼芽生根的最适培养基.研究结果可为泡桐的基因工程操作和阐明丛枝病发生机理奠定基础.
Establishment of plantlet regeneration system with leaves, petioles and nodal segments of Paulownia tomentosa×Paulownia fortunei with witches' broom was studied in this paper. The results indicated that the highest shoot induction rates of the leaves, petioles and nodal segments on the following combined media, MS + NAA 0. 3 mg·L^-1+ 6-BA 12mg·L^-1,MS+ NAA0.3 mg·L^-1+6-BA8 mg·L^-1 and MS+ NAA0.3 mg·L^-1 + 6-BA 12 mg·L^-1 were respectively 47.78% ,21.11% and 42.22% .1/2MS free NAA and IBA might be the optimal medium for shoot rooting induction. These results might lay solid foundation for the manipulation of gene engineering of P. tomentosa × P .fortunei with witches' broom.
