摘要
我们首先报道了RAPD-PCR技术研究微小牛蜱DNA的多态性。本文则用3个引物"Pz"(CGGCCCGGTA)、"Pw"(CGGCCCCGGT)和"Py"(CTGCAGGACA)对亚洲璃眼蜱基因组DNA作RAPD-PCR,均能扩增出多态性DNA图谱;各引物扩增的DNA片段的分子大小是:"pz"为1380、1230、955、912、871、794、741、668、646、562、537、442;"pw"为955、724、668、562、537、457、279;"Py"为1072、912、832、794、612、582、562、537、422、385。RAPD-PCR技术简便、快速,并能在DNA水平上揭示其差异和进化关系,可发展成为理想的蜱类分类学技术之一。
We reported the study on polymorphism DNA of Boophilus microplus with RAPD-PCR technique in the first instance.Here,we amplified the genomic polymorphism DNA maps of Hyaloma asiaticum asiaticum with three primers-“Pz” (CGGCCCGGTA),“Pw”(CGGCCCCGGT) and“Py”(CTGGAGGACT).The molecular weights (bp) of the DNA bands for each map were:“Pz”: 1380,1230,955,912,871,794,741,668,646,562, 537,442; 'Pw':955,724,668,562,537,457,279; 'Py':1072,912,832,794,612,582,562,537,422,385. The RAPD-PCR for its simplicity, rapidness,and being able to reveal difference and evolutional relationships at the level of DNA,could be developed into an ideal taxonomic method for Acarology.
