摘要
研究了用微柱高效液相色谱法测定丹参中的4种有效成分(丹参素,原儿茶酸,原儿茶醛,丹酚酸B)的方法.丹参样品中的几种有效成分用体积分数40%的甲醇超声振荡提取,然后以Waters XterraTM RP18(1.0×50 mm,2.5μm)微柱为固定相,甲醇和体积分数1%的HAc溶液梯度洗脱为流动相分离,在该色谱条件下,丹参中的4种有效成分在4.0min内可达到基线分离;用紫外二极管矩阵检测器检测.方法标准回收率为97%~102%,相对标准偏差为1.6%~2.3%.用此方法可测定几种丹参样品中的有效成分.
A microcolumn high performance liquid chro- matography (HPLC) method for the determination of four effective components (danshensu, protocatechuic aldehyde, protocatechuic acid, salviamolic acid B) in Salvia Miltiorrhiza was studied. The four effective components were extracted from samples with 40% methanol by ul- trasonic extraction for 45 rain. Then the four effective components was separated on a Waters Xterra^TM RP18 (1.0 - 50 mm, 2.5 μm) chromatographic microcolumn with methanol and 1% acetic acid solution as mobile phase using gradient elution at a flow-rate of 0. 15 mL/min, and monitored with a photodiode array detector at 280 nm. The recoveries of the flavonoids are 95 % 102%, and the relative standard deviations are 1.2% - 2.3 % . This method was applied to the determination of danshensu, protocatechuic aldehyde, protocatechuic acid and salviamolic acid B in Salvia Miltiorrhiza with good results.
