摘要
目的:观察γ-干扰素(IFN-γ)对血管内皮细胞(EC)的作用及其对肿瘤坏死因子-α(TNFα)表达的影响。方法:体外培养EC,MTT法检测IFN-γ(0、20、40、80μg/L)对EC生长的直接影响,半定量逆转录-多聚酶链反应检测不同浓度的IFN-γ对EC凋亡相关基因TNFαmRNA的表达的影响。结果:IFN-γ可抑制体外培养的EC生长(P<0.01),且具有剂量依赖;IFN-γ能明显上调EC的TNFα表达。结论:IFN-γ可能通过上调EC中TNFα的表达而诱导EC凋亡。
Objective:To explore the role of interferon-γ(IFN-γ) in inducing the apoptosis of vascular endothelial cells(EC) in vitro and its influence on the expression of TNFα. Methods:The EC cells were cultured in vitro with the condition media containing varied concentration of IFN-γ(0,20,40,80μg/L) for 24 hours,and then the total RNA was extracted to analyze the mRNA level of TNFα in EC by RT-PCR. The influence of IFN-γ in inducing the apoptosis on EC was analyzed by MTT method. Results: IFN-γ could induce the apoptosis of EC in a dose-dew.ndent manner and it could upregulate the expression of TNFα mRNA level in EC. Conclusions: IFN-γ can induce the apoptosis of EC in vitro by up-regulating the expression of TNFα.
