胰腺炎相关腹水对大鼠胃肠动力的影响 被引量:10

摘要 目的探讨急性胰腺炎相关腹水(PAAF)对离体胃肠平滑肌运动的影响。方法以5%牛磺胆酸钠溶液逆行注入胰胆管制胰腺炎(AP)模型,24h后收集腹腔液,低温离心,取上清液作为胰腺炎相关腹水。用Maclab数据分析系统,描记离体肠段的运动。观察20%、40%浓度的胰腺炎相关腹水对离体空肠段运动的影响,利用逆转录-聚合酶链反应(RT-PCR)法检测空肠组织iNOSmRNA的表达。结果PAAF可引起离体空肠运动峰值频率降低和平均振幅减弱,10-4mol/L的Nω"硝基"L"精氨酸(L"NNA)可逆转20%PAAF对离体肠管运动频率和振幅的影响。正常空肠组织iNOSmRNA不表达或低水平表达,20%腹水组iNOSmRNA为1.07±0.11;40%腹水组为1.15±0.07,呈高表达。结论急性PAAF抑制胃肠蠕动是通过降低胃肠平滑肌的运动频率和振幅,腹水可能通过增加胃肠组织内iNOS活性产生大量NO而抑制肠动力。 Objective To investigate the effect of pancreatitis-associated ascite fluid on gastrointestinal motility. Methods AP was produced with 5% sodium taurocholate by retrograde pancreatic-injection. At 24hr after the induction of pancreatitis, the rats were sacrificed and the peritoneal exudates were aseptically collected followed by centrifugation at 40C. The surpernatants from the rats were designated as pancreatitis-associated ascitic fluids(PAAF). We completed directly A/D change and achieved the computer real-time recording by combining the traditional recording device with Maclab/4e date recording and analysis systems. The effect of 20% PAAF on the motility of the rat jejunum section in vitro was studied and the jejunum iNOS mRNA expression was assayed by reverse transcription-PCR. Results The motility frequency and motility amplitude of the rat jejunum section were inhibited significantly by PAAF (P〈0.05; P〈0.01), 10^-4nol/L L-NNA could inhibit the effect of 20% PAAF on rat jejunum. There was over expression of the iNOSmRNA in rat jejunum (P〈 0.01). Conclusions The motility frequency and motility amplitude of gastrointestinal can be inhibited by PAAF in vitro. L-NNA can inhibit the effect of PAAF on rat jejunum; Our results suggest that PAAF play a role in the gastrointestinal dysmotility during acute pancreatitis, at least in part through the activation of iNOS.
