摘要
目的探讨金龙胶囊对肿瘤复发/转移的作用及其机制。方法利用小鼠宫颈癌U14复发转移动物模型,观察金龙胶囊对肿瘤复发/转移的影响;采用体外培养肿瘤细胞,观察金龙胶囊对HL60细胞形态变化、四唑氮蓝(NBT)还原能力及吞噬能力的影响;细胞化学法观察MGC803细胞PAS阳性物质的表达情况,荧光染料传递观察细胞间通讯改变;免疫组化观察DCS细胞层黏连蛋白(LN)表达的变化。结果金龙胶囊对U14复发瘤的瘤重抑制率为54.8%～66.3%,肺转移抑制率为50%～54%。金龙胶囊作用后HL60细胞NBT还原能力、吞噬能力增加,形态上成熟细胞增多;MGC803细胞PAS阳性物质增多,细胞间小分子染料传递增强,DCS细胞LN表达增加。结论鲜中药金龙胶囊可显著抑制肿瘤的复发/转移,作用机制包括促进肿瘤细胞分化和促进肿瘤细胞间通讯。
Objective To explore the effects and the mechanism of fresh TCM recipe on tumor recurrence. Methods The effects and the mechanism of fresh TCM recipe on recurrence rate and metastatic rate of animal model of tumor recurrence were observed. The morphological changes, NBT reduction, phagocytosis, dye-transfer, laminin（LN） and PAS expression were used to evaluate the effects of fresh TCM on the induction of tumor cell differentiation. Results The inhibition rate of fresh TCM recipe on U14 recurrent tumor was between 54.8%～66.3%. The inhibition rate of fresh TCM recipe on lung metastasis of U14 was between 50%～54%. After treatment with fresh TCM recipe the NBT reduction ability, the phagocytic ability of HL60 cells was increased and morphologically more mature than before treatment. There were more PAS positive substances in MGC803 cells after treatment and l＇uciffer yellow transferred faster and further between the MGC803 cells. The expression of LN in DCS cells was increased after fresh TCM recipe treatment. Conclusions The fresh TCM recipe shows marked inhibition of tumor recurrence and metastasis. The possible Mechanism includes differentiation induction and inter cellular communication promotion.
