破骨细胞分化因子及其受体和反意受体在骨巨细胞瘤中的表达及意义

摘要 目的:检测破骨细胞分化因子(ODF)及其受体(RANK)和反意受体-护骨因子(OPG)在骨巨细胞瘤(GCT)组织中的表达,探讨其与GCT中多核巨细胞形成J、affe分级和临床复发的关系。方法:用逆转录聚合酶链反应(RT-PCR)法检测5例GCT组织中ODF、OPG、RANK mRNA的表达,用免疫组织化学法检测44例GCT组织中ODF蛋白的表达,统计分析其表达与Jaffe分级和临床复发的关系。结果:GCT组织中ODF、OPG、RANK mRNA均有表达,且ODF表达丰富;GCT组织单核基质细胞和多核巨细胞中均有ODF蛋白的表达,但以单核基质细胞为主,Ⅰ级单核基质细胞中ODF蛋白的表达明显高于Ⅱ、Ⅲ级,ODF蛋白的表达与复发无关。结论:GCT组织中ODF的表达与多核巨细胞的形成及Jaffe分级有关。 Objective To detect the expression of osteoclast differentiation factor(ODF) and its receptor(RANK) and decoy receptor-osteoprotegerin(OPG) in giant cell tumor of bone;to study its effect on the formation of muhinucleated giant cells and the relationship between the Jaffe grade and prognosis. Methods Using RT-PCR ODF, OPG, RANK mRNA were detected in 5 cases of giant cell tumors. Using immunohistochemical analysis the expression of ODF at protein level was evaluated in 44 cases of GCT tissues, their correlations with the Jaffe grade and clinical recurrence were analyzed statistically. Results ODF, OPG, RANK mRNA were all expressed in GCT tissues, among which ODF was expressed in abundance; ODF protein were expressed in the multinucleated giant cells (MGC)and the mononuclear cells (MC) in GCT tissues. Most of MC cells expressed ODF. The expressing rate of ODF protein in MC in cases of grade I was significantly higher than that in cases of grade Ⅱ and Ⅲ ; The expressing rate of ODF in MC had no correlation with recurrence. Conclusion The expression of ODF correlate with the formation of muhinucleated giant cell and Jaffe stage.
