术前术中持续输注谷氨酰胺对大鼠肝脏缺血再灌注损伤的影响 被引量:2

Effect of Continuous Glutamine Infusion before and during Operation on Hepatic Ischemia Reperfusion Injury in Rats

摘要 目的研究术前术中持续输注谷氨酰胺对大鼠肝脏缺血再灌注损伤的影响。方法 16只雄性Wistar大鼠随机分为实验组与对照组,每组8只,2组均行右侧颈外静脉插管,实验组输入3%丙氨酰-谷氨酰胺二肽(AlaGln),对照组输入生理盐水,连续输注3天,直到缺血再灌注结束。Pringle's法行肝缺血40分钟,再灌注1小时后检测门静脉血中内毒素(ET)、主动脉血中谷丙转氨酶(ALT)、谷草转氨酶(AST)、乳酸脱氢酶(LDH)及肝组织细胞间黏附分子-1(ICAM-1)的表达。结果实验组ET水平(0.3229±0.0756)EU/ml显著高于对照组(0.2021±0.0495)EU/ml(P<0.01);实验组ALT水平(407.29±84.27)U/L显著高于对照组(312.67±66.48)U/L,(P<0.05);AST、LDH两组比较差异无显著性。实验组肝组织ICAM-1表达较对照组明显增强(P<0.05)。结论术前术中持续输注谷氨酰胺对大鼠肝脏缺血再灌注损伤可能是有害的。 To study the effect of continuous glutamine infusion before and during operation on hepatic ischemia/reperfusion injury in rats. Methods Sixteen male Wistar rats were randomly divided into experimental group and control group. Rats in both groups were cannulated in right external jugular vein. In experimental group, alanyl-glutamine (Ala-Gln) were continuously infused for 3 days till the end of ischemia/reperfusion. Normal saline was used in the control group. Hepatic ischemia/ reperfusion injury model with portal congestion was established according to Pringle's maneuver for 40 minutes. After one hour of reperfusion of ET (endotoxin) in portal vein, alanine aminotransferase (ALT), aspartate aminotransferase (AST), and lactic dehydrogenase (LDH) levels in abdominal aorta and ICAM-1 (intercellular adhesion molecule-l) in liver tissue were examined. Results The ET levels of experimental group and control group were (0.3229 ± 0.0756) EU/ml and (0.2021 ± 0.0495) EU/ml (P〈0.01); ALT levels were (407.29±84.27) U/1 and (312.67±66.48) U/1 (P〈 0.05). There were no significant differences on AST and LDH levels between two groups (P 〉 0.05); ICAM-1 expression in experimental group was significantly higher than that in control group (P 〈 0.05). Conclusions Continuous glutamine infusion before and during operation may be harmful on hepatic ischemia/reperfusion injury.
