天然抗氧化剂对抗PC12细胞氧化损伤的实验研究 被引量:3

摘要 目的:研究五味子乙素(SchB)对H2O2诱导的PC12细胞氧化损伤的保护作用及作用机制。方法:SchB与PC12细胞预培养24h后,加入H2O2处理,在倒置显微镜下观察细胞的形态改变,采用MTT法测定细胞生存率,采用单细胞凝胶电泳法测定DNA单链损伤,采用荧光法测定细胞内的谷胱甘肽(GSH)含量。结果:H2O2处理后细胞生存率明显降低,细胞内DNA单链损伤明显升高,而GSH含量明显降低。经SchB预培养的细胞生存率明显升高,细胞内DNA单链损伤明显减轻,GSH含量明显升高,其作用呈剂量依赖关系。结论:SchB对PC12细胞有明显的保护作用,其作用机制可能与提高GSH抗氧化系统的能力有关。 Objective: To study the protective effects and mechanism of Schisandrin B (Sch B) on oxidative damage induced by H2O2 in PC12 cells,Methods: The ceUs were treated with H2O2 after incubated with SCh B for 24 hours. The morphology of the cells was observed by microscope and the cell viability was measured by MTI method, DNA single breaks by Comet assay and the GSH contents by fluorescent method. Results: The cell viability and the GSH contents were decreased obviously, DNA damage were increased. Sch B increased the cell viability markedly, protected the cells from H2O2 - induced DNA damage and increased the contents of GSH in a dose-dependent manner. Conclusion:It is shown that SCh B can protect the PC12 cells against oxidative injury by enhancing the ability of antioxidant.
