摘要
采用PCRR-FLPs技术,对香猪、上海白猪、大约克夏猪生长激素基因-119~+715区域共834bp片段的扩增产物分别用ApaI、DraI、MspI3种限制性内切酶检测酶切位点的多态性。结果显示:ApaI酶切时,3个品种的猪都产生了3种基因型(AA、BB和AB),BB基因型的频率以大约克夏猪最高,AB基因型的频率以香猪最高,3个猪种间基因型频率、等位基因频率的差异不显著(P>0.05);DraI酶切时,各品种猪均未呈现酶切位点的多态性;MspI酶切时,仅大约克夏猪表现出2种基因型(CC、CD),香猪、上海白猪未产生酶切位点多态性。结论:AB基因型的频率以香猪为最高,它可能是小型猪的有利基因型;在香猪和上海白猪中DraI和MspI酶切位点的多态性比较贫乏。
Apa Ⅰ, Dra Ⅰ and Msp Ⅰ polymorphisms of a 834bp ( - 119 - + 715) fragment of porcine growth hormone (pGH) gene were analyzed by PCR- RFLPs in Xiang Pig, Shanghai White and Large Yorkshire. The results showed that there were three genotypes (AA, BB, AB) in every porcine breed when the PCR products were cut by Apa Ⅰ, BB genotype frequency of Large Yorkshire was highest in three breeds and AB genotype frequency of Xiang Pig was highest in three breeds, and the difference of genotype frequencies and allele frequencies in three breeds was not significant ( P〉 0. 05). No Dra Ⅰ polymorphism of pGH gene was seen in these porcine breeds. When the PCR products were cut by Msp Ⅰ, there were two genotypes(CC, CD) in Large Yorkshire breed and three were not polymorphism of pGH gene in Xiang Pig and Shanghai White. Conclusion: It is possible that AB genotype is the helpful genotype of miniature pig for its highest genotype frequency in Xiang Pig. Dra Ⅰ and Msp Ⅰ restriction enzyme site polymorphisms of pGH gene are poor in Xiang Pig and Shanghai White.
