重组人干扰素-α2a(IFN-α2a)的圆二色谱分析 被引量:5

摘要 目的:研究测量温度和蛋白质浓度对重组人干扰素-α 2a(rhIFN-α 2a)圆二色谱的影响,进而研究对 rhIFN-α 2a 二级结构比例的影响。方法:分别研究了测量温度和蛋白质浓度对 rhIFN-α 2a 圆二色谱的影响,另外还对不同厂家、不同批次的4批 rhIFN-α 2a 的圆二色谱进行了比较。结果:测量温度在60~95℃之间,rhIFN-α 2a 圆二色谱的形状发生改变,三级结构遭到破坏,二级结构发生改变;在5~60℃温度区间,二级结构相对稳定。rhIFN-α2a 的各二级结构比例在1.5~0.006nag·mL^(-1)范围内相对稳定。不同厂家、不同批次的4批 rhIFN-α 2a 的圆二色谱形状基本一致,4种二级结构的比例相近。结论:在一定的温度和浓度范围内,rhIFN-α 2a 的圆二色谱相对稳定,且不同批次的测量结果具有很好的一致性。圆二色谱可用于蛋白质类药物的质量控制。 Objective:To study the effeαs of temperature and concentration on circular dichroism (CD) speαra of recombinant human interferon alpha 2a ( rhIFN - α 2a) and the estimation of protein second struαure proportion. Method: CD speαra of rhIFN -α 2a was determined in different temperature and concentration and the second struαure proportion of rhIFN - α 2a was estimated by Self - Consistent Methods. In addition, the CD speαra of four different batches rhIFN - α 2a were compared each other. Result:The speαra and second struαure proportion of rhIFN - ot 2a were stable from 5 ℃ to 60 ℃ and changed from 60 ℃ to 95 ℃. The second struture proportion of rhIFN - α 2a was stable with the decreasing of concentration from 1.5 mg · mL^ - 1 to 0. 006 mg · mL^ - 1. The comparison of rhIFN - α 2a from different resources and different batches showed that the speαra and second struαure proportion were consistent with each other. Conclusion:The speαra and second struαure proportion of rhIFN - α 2a were stable in the special range of temperature and concentration. The speαrum and second struαure proportion of rhIFN -α 2a from different resources and different batches were consistent with each other. CD can be applied to the oualitv control of bio - pharmaceutics.
