摘要
目的观察苦参碱对UUO大鼠模型肾小管间质CTGF和-αSMA表达水平的影响,初步探讨其对小管间质纤维化的作用机制。方法实验采用UUO肾间质纤维化模型作为研究模型,分为正常组、假UUO组、手术组(UUO组)、福辛普利治疗组(F组)、苦参碱大剂量治疗组(苦大组)和苦参碱小剂量治疗组(苦小组)。运用免疫组化技术半定量分析实验第7,14天各组的CTGF和-αSMA表达水平。结果UUO组和治疗组的CTGF和-αSMA的表达水平在UUO模型建立后的第7,14天均显著高于正常组和假UUO组(P<0.05),但3个治疗组的表达水平明显低于UUO组(P<0.05);CTGF和-αSMA在苦大组与F组间无统计学差异(P>0.05)。结论苦参碱可以降低肾小管间质CTGF和-αSMA的表达水平,延缓肾小管间质纤维化的进程。
Objective Our aim of the study is to observe the effects of the matrine on the renal tubulointerstitium, to explore the mechanisms of its effects. Methods 70 male SD rats were randomly divided into 5 groups:normal group, shame UUO group, UUO group. UUO group treated with the fusinopril ( F group ), UUO group treated with the large dose of matrine ( large matrine ) and UUO group treated with the small dose of matrine ( small matrine) . The expression levels of CTGF and α-SMA in all the groups were determined by immunohistochemistry at 7,14 days. Results The expression levels of the CTGF and α-SMA in the UUO group and treatment groups were significantly increased compared to the normal group and shame UUO group ( P 〈 0.05), but the expression levels in the treatment groups were lower than that in the UUO group (P〈0.05) ;there was no significant difference between the large matrine group and the fusinopril group (P〉0.05). Conclusion Matrine could decrease the expression levels of the CTGF and α-SMA in the renal tubulointerstitium, delay the progression of the renal tubulointerstitial fibrosis.
