摘要
目的:探讨支气管哮喘患儿气道粘膜上皮细胞是否表达基质金属蛋白酶,以及它们与患儿病情的关系,并探讨应用糖皮质激素对其影响。方法:选取50例支气管哮喘患儿及来我院的需要做支气管黏膜活检的非哮喘患儿20例,进行纤维支气管镜黏膜活检,对活检组织做TIMP-1(组织抑制因子-1)与MMP-9(胶质基质金属蛋白酶)、MMP-3(硫铜基质金属蛋白酶)免疫组化染色;同时对患儿进行组胺激发实验。结果:所有支气管哮喘患者气道黏膜上皮MMP-9、MMP-3免疫组化染色均为阳性,相关分析显示TIMP/MMP-9比值与病情的轻重呈正相关,应用糖皮质激素可以改变支气管黏膜的TIMP-1与MMP-9、MMP-3平衡。结论:支气管哮喘患者气道黏膜上皮表达MMP-9、MMP-3、TIMP-1与MMP-9、MMP-3的不平衡也许是导致支气管哮喘发生的原因,应用糖皮质激素可以调节这个平衡.
Objectlve To investgate the expression of matrix metalloproteinases in bronchial biopy mucosal tissue and its relation to state of the illness.discoving the effect of glucorticosteroid Method 50 paitents with bronchial asthma with and 20 Children with non -asthma child who need bronchial mucosal tissue biopy were recruited, the biopy was carried out to all pafients.immunohistochemistry was employed for TIMP-1,MMP-9,MMP-3.meanwhile,excitated test with histamin was explored. Results immunohistochemistry for TIMP-1,MMP-9,MMP-3 at bronchial biopy mucosal tissue of asthma child showed positiveness correlation analyis showed that the ratio of TIMP/MMP-9 was positively correlated with grade of illness. Glucorticosteroid can regulate the balance of the expression of TIMP-1,MMP-9,MMP-3 at bronchial biopy mucosal tissue. Conclusion The unbalance of TIMP-1,MMP-9, MMP-3 expressed at bronchial biopy mucosal tissue of patients, may contribute the pathogeny of bronchial asthma,glucorticosteroid can regulate the balance.
