摘要
目的:建立一种可测定炎可宁糖衣片中盐酸小檗碱和黄芩苷含量的RP-HPLC方法。方法:采用Hypersil-BDSC18(200mm×4.6mm5μm)为色谱柱;流动相:黄芩苷为乙腈-0.1%磷酸溶液(22:78);盐酸小檗碱乙腈-磷酸盐缓冲液〔0.05mol/L磷酸二氢钾和0.05mol/L庚烷磺酸钠(1:1)含0.2%三乙胺〕(40:60);盐酸小檗碱和黄芩苷的检测波长分别为347nm与278nm;柱温:30℃;进样量均为20μL;流速均为1.0mL/min;峰面积外标法定量。结果:本法盐酸小檗碱在16.0~128μg·mL-1,黄芩苷在40.2~321μg·mL-1范围内两者均有良好的线性关系(r=0.9999),黄芩苷和盐酸小檗碱的平均回收率与RSD%分别为99.1%,(RSD)0.28%和99.7%,(RSD)1.03%。结论:方法简便,快速,准确可靠,可作为该制剂的质控方法。
Objective : To establish an RP - HPLC method for determining the content of berberine and scutellarin in Yankening tablets. Methods :The berberine and scutellarin were analyzed on a Hypersil - BDSC18(200mm × 4.6mm 5μm) column, and detected respectively at 347nm and 278nm, The temperature of column was at 30℃, The samples injected 20μL and the flow rate was 1.0mL'min^-1,Acetonitrile- 0.1% phosphric acid(22: 78) was used as a mobile phase for scutellarin and acetonitrile - phosphate buffer(0.05mol/L potassium dlhydrogen phosphate- 0.05mol/L sodium heptanesulfonate 1 : 1, contain 0.2 % triethylamine)(40:60) was used as a mobile phase for berberine, External standard method was used. Results: The linear ranges of berberine and scutellarin were 16.0 - 128μg · mL^-1 ( r = 0.9999) and 40.2 - 32μg · mL^-1 ( r = 0.9999), and the average recovery were 99.8 % ; (RSD)1.03% and 99.1% ; (RSD)0.28% respectively, Conclusion :The method is simple, rapid and accurate for quality control of the preparation.
