摘要
目的:研究牛乳铁蛋白(BLF)的质量控制。方法:采用高效毛细管电泳法(HPCE)对牛乳铁蛋白的含量进行测定。结果:牛乳铁蛋白的浓度在100.0-600.0μg·ml-1与其峰面积成良好的线形关系,r=0.999 5。结论:该法操作简便,精密度好,结果准确可靠,可作为牛乳铁蛋白质量标准的重要指标。
Objective: To study the quality control of bovine lactoferrin by HPCE. Method: The method employed a 33 mmol· L^-1 potassium dihydrogen phosphate- 10 mmol·L^-1 phosphoric acid solution (pH 2.50) as buffer solution at an electrophoresis voltage of 25 kV. The detection wavelength was 214 nm and the uncoated fused silica capillary column was 50μm × 51 cm. Before sample injection, the capillary was rinsed with 0.1 mol·l^ - 1 sodium hydroxide solution, highly pure water and buffer solution for 5 min, 3 min and 3 min, respectively. Pressure injection was used for 6.9 kPa × 5 s. The temperature was at 25 ℃. Result: A good linearity was showed in the concentration range of 100.0-600.0μg· ml^- 1 for bovine lactoferrin( r = 0.999 5 ). Conclusion: The method is simple and accurate. It can be used for quality control of bovine lactoferrin.
