摘要
目的:建立高效液相色谱法测定海棠合剂中雷公藤内酯醇含量的方法.方法:样品经以氯仿为洗脱剂,中性氧化铝柱层析的方法进行分离、提纯后,采用高效液相色谱法,SHIM-PACK VPODSC18(150 mm × 4.6 mm,5 μm)色谱柱,以乙腈-水(23∶77)作为流动相,在检测波长220 nm处测定.结果:在 2.74 ～96 mg·L -1 范围内,峰面积与浓度线性关系良好,r= 0.999 9 ;平均加样回收率 96.99% ,RSD为 1.79% (n=6).结论:方法灵敏、结果准确,适用于海棠合剂中雷公藤内酯醇的含量测定.
OBJECTIVE To develop a method for determination of triptolide in Haitang mixtures. METHODS An HPLC method was carried out using SHIM-PACK VPODSC18（150 mm ×4. 6 mm,5μm）column and a mobile phase consisting of aeetonitrile-water （23：77）. The detection wavelength was 220mm, The flow rate was 1.0 mL·min^-1. RESULTS The calibration curves was linear in the concentration range of 2. 74 - 96mg·L^-1 （r= 0. 999 9,n = 6）. The average recovery was 96. 99%（RSD= 1.79%）. CONCLUSION The method is accurate,sensitive. It is suitable for the determination of triptolide and its preparations.
