摘要
目的探讨南蛇藤素对肿瘤血管抑制作用。方法通过建立动物肿瘤模型,对南蛇藤素予以实验治疗,结合免疫组化,检测其抑瘤率和肿瘤内血管密度,以及瘤体VEGF和bFGF的表达的改变。结果南蛇藤素具有明显的抑制S180肉瘤作用,其3mg/kg剂量组抑瘤率达46.86%,与5Fu联用抑瘤率达60.2%,联合给药组和南蛇藤素3mg/kg剂量组的微血管密度、VEGF、bFGF的表达均明显低于对照组间和5Fu组,差异有显著性(P<0.01)。结论南蛇藤素对小鼠S180肉瘤血管有一定的抑制作用,降低VEGF、bFGF的表达可能是其抑制肿瘤血管形成的机制之一。
Objective To investigate the effect of inhibiting angtogenestsDy celastrol inhibitory rate and the vascular density were tested in animal tumor model with experimental treatment. The expression of VEGF and bFGF were measured by immunohistochemistry. Results The tumor inhibi tory rate was 46. 86% in celastrol(3mg/kg) ,and was 60. 2% in combined group. The intratumoral microvessel density(MVD),expression of VEGF and bFGF in combined group and celastrol(3mg/kg) group were lower than the control and 5-Fu group(P〈0.01). Conclusion Celastrol have inhibitory effect on S180. sarcoma and strong inhibitory effects on angiogenesis. Its mechanism may be associated with the down-regulating of VEGF and bFGF expression.
